1950 में उन्हें वैटिकन से उनके मिशनरीज ऑफ चैरिटी की स्वीकृति मिली. उन्होंने कोढ़ रोगियों के लिए विशेष कार्य किए जिन्हें समाज से एक तरह से बाहर ही कर दिया जाता था. इसके बाद वे गरीबों की झोपड़ पट्टी में खुद जाकर रहने लगीं इस दौरान उन्हें किसी तरह का सहयोग नहीं मिला यहां तक कि खुद को जिंदा रखने के लिए उन्हें भीख तक मांगनी पड़ी और लोगों ने संदेह के नजरिए से देखते हुए उन्हें नजरअंदाज किया. उन्होंने बंगाल में मिशिनरियों के सेवाकार्य की काहनियां भी सुनी जिससे भारत आने के विचार उनके मन में पनपने लगे. एक्नेस साल 1929 में भारत पहुंचीं और दार्जलिंग में रह कर बंगाली सीखी और अपने कॉन्वेंट के पास शिक्षण कार्य भी शुरु किया. इसी समय जब दार्जलिंग जा रहीं थी, तब उनके मुताबिक उनके अंदर से आवाज आई और उन्हें एहसास हुआ कि ईश्वर उनसे गरीबों की सेवा करने का आदेश दे रहे हैं.
